Identification of 16S rRNA gene sequences from thermophilic bacteria that showed activity of hydrolytic enzyme exo-type on inulin substrate

Abstract

Potential sources of thermostable enzymes are thermophilic bacteria. In the research, nucleotide base sequences of 16S rRNA gene from thermophilic bacteria have been identified. The bacteria showed activity of hydrolytic enzyme exo-type on inulin substrate. The bacteria were isolated from 'Batu Bajanjang' hot spring, Solok (West Sumatra) that labeled as BB3 isolate. Gene of 16S rRNA fragment was isolated using PCR technique with BactF1 and UniB1 primers. The nucleotide base sequences of 16S rRNA gene were determined using the dideoxy-Sanger method. Analysis of the gene fragment (1348 bp) from BB3 isolate indicated as Fictibacillus gelatini or Bacillus gelatini species. Activity of extracellular enzyme indicated exo-type action based on analysis of the inulin hydrolysis end products using Thin Layer Chromatography (TLC). So, the enzyme is the potential to produce high fructose syrup from inulin. © 2023 Author(s).
